Well, there is a set of tricks or additional information that you can use to your advantage to know more. Although it is impossible to be sure if another user has read your messages or not, you can know what his status is, if he is active, or if he is not. Normally, if he's active, he should have read the message.
By default, the user's online status is indicated by a green dot next to their avatar . When you notice a yellow moon icon, it means that you are Inactive, that is, online but a bit busy. Finally, a red icon will mean that he does not want to be disturbed and will not be notified of messages.
